Properly targeted ES cells containing a homologous recombination event were selected, and transfected with a Cre expressing vector to excise the resistance cassette. Targeted ES cells were then injected into C57BL/6 blastocysts. Chimeric offspring were bred with C57BL/6 mice to yield mice heterozygous for the mutant allele.
