Raf-1 was targeted by homologous recombination with the targeting vector. Correctly targeted ES cells were transfected with a Cre expressing vector to excise the resistance cassette, before injection into C57BL/6 blastocysts. Chimeric offspring were bred with C57BL/6 mice to yield mice heterozygous for the mutant allele.
