  • Description: Anti-RNA polymerase II (RNAPII), clone CTD 4H8 is mouse monoclonal antibody. CTD 4H8 recognises both hypo- and hyperphosphorylated RNAPII. CTD 4H8 is known to be reactive with the C-terminal domain (CTD) in humans and saccharomyces cerevisiae.

    RNAPII is a multi-subunit enzyme responsible for the transcription of protein-coding genes. The C-terminal domain (CTD) is a highly conserved domain unqiue to RNAPII, coordinating factors that regulate transcription initiation, elongation and mRNA processing. Mutations that directly affect transcription by RNA polymerases rank among the most central mediators of malignant transformation.

    The immunogen is a peptide containing 10 repeats of synthetic peptide YSPTSPS using chemically synthesised phospho-Ser5.

  • Target background: RNAPII is an essential component of the RNAP II transcription elongation complex. Recruitment of RNA processing factors to the elongation complex is coordinated by the phosphorylation of Serine-2 and Serine-5 residues within the C-terminal repeat (YSPTSPS) domain of the largest subunit of RNAPII. Mutations that directly affect transcription by RNA polymerases rank among the most central mediators of malignant transformation. CTD4H8 can also be used in chromatin immunoprecipitation (ChIP) assays.
