Target Details

  • Target: Haemoglobin Scavenger Receptor (M130; CD163)
  • Target background: Anti-haemoglobin scavenger receptor labels the majority of monocytes and tissue macrophages. It recognises a membrane glycoprotein with molecular weight 130,000 (reduced) and 110,000 (unreduced). The glycoprotein antigen is expressed exclusively on the cell surface of human monocytes and macrophages, which evolves predominantly in the late phase of inflammation. The glycoprotein GHI/61 binds to CD163, a transmembrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It is a receptor for the haemoglobin-haptoglobin complex. CD163 expression on monocytes and macrophages changes as these cells differentiate, as such GHI/61 can detect levels of differentiated monocytes and macrophages. CD163 has high affinity for the haemoglobin-haptoglobin complex and upon ligand binding, leads to the production of IL-6 and IL-10. GHI/61 monoclonal antibody was created to label monocytes and macrophage with increased specificity, given similar antibodies frequently label lymphocytes etc., or only recognise a particular subset of macrophages.
